Ames test

Ames test는 bacterial test organism을 이용하여 mutagen을 가려내는, 간단하면서도 비용이 많이 들지 않는 방법이다. Test organism은 histidine-negative(his-)인 Salmonella typhimurium의 영양요구체(auxotroph) 균주이다. 이 균주는 back-mutation이 일어나 his+로 바뀌지 않는 한 histidine이 없는 배지에서 자라지 못할 것이다. Chemical의 mutagenic effect는 종종 organism의 enzymatic pathway에 의해 영향을 받아 nonmutagen이 mutagen으로 전환되거나 혹은 반대로 mutagen이 nonmutagen으로 전환되기도 하는데, 포유류에서는 이러한 toxification이나 detoxification이 간(liver)에서 일어난다. 그래서 Ames test에서는 bacterial system이 mammalian test system에 더 근접하도록 하기 위해 activating enzyme의 source로서 liver homogenate S-9를 넣어준다. Spot method에 의한 Ames test에서는 test organism, S-9, molten agar와 미량의 histidine - bacteria로 하여금 mutation에 필요한 몇번의 cell division을 할 수 있도록 하기 위해 넣어주는 것이다 - 을 섞어 minimal agar plate에 붓고, test chemical로 적신 disc를 test plate의 가운데에 놓는다…
