B. Processing and Formulation Condition

1) 온도

- 대부분의 단백질은 온도 증가와 더불어 가역적으로 denaturation
- 온도의 계속적 증가로 denaturation촉진되고 cooling으로도 reverse되지 않음
- 어떤 경우에 original native structure 와 다른 새로운 구조로 unfold되고 refold됨
- 온도를 낮춘 경우에도 native structure로의 refolding 방해하는 높은 kinetic energy barrier
때문에 protein은 이런 misfolded된 상태로 존재
- 낮은 온도에의 노출은 cold denaturation : reverse enthalpic & entropic effect로 일어남

2) pH

- formulation buffer의 pH는 화학적으로 amino a챵 잔기 바꾸는 protein solubility, folding,
반응속도에 심각한 충격을 줄 수 있다.

3) Ionic Strength 이온강도

- counterion의 농도는 분자에서 정전기적인력을 매개하는 중요한 특성이고 protein의
solubility뿐만 아니라 stability에 큰 효과를 가진다.
- 전해질의 Debye-Huckel theory : 정전기적 인력에 counterion농도의 효과
